Serine Protease Inhibition Kits

Three new kits are offered for the study of inhibition of serine proteases in whole cells. Red FLISP, SR101-FCK; Red FLISP, SR101-LCK; and Green FLISP, FAM-FCK (FLISP is an acronym for fluorescently labeled inhibitors of serine proteases) work in whole cell assays with protocols for use in flow and laser scanning cytometry as well as in fluorescent microplate readers. Using these reagents, novel serine proteases involved in protein degradation during apoptosis in T24, HL-60 and other cells have been detected by simultaneous inhibition and covalent binding of the well-known sulforhodamine 101 or FAM labels to the proteases. FLISP reagents do not label caspases, so activation of caspases can be measured simultaneously with Green or Red FLICA probes. FLISP probes can be used to study antagonists as well as agonists of the as-yet uncharacterized serine proteases important to the apoptotic process.
