Isolation Of Human Regulatory T Cells

The purification of CD4+CD25+bright regulatory T cells (TReg) has historically been a laborious, multi-step process, and is often complicated by contamination with activated (CD4+CD25+dim) T cells. Now a rapid, simple method for TReg isolation is available. RosetteSep CD4+ T cell enrichment directly from whole blood is followed by manual (EasySep) or fully automated (RoboSep) positive magnetic selection of only high-expressing CD25+bright cells. TReg purities of up to 90% can be achieved in 2-3 hours. The isolated cells express the FOXP3 transcription factor, which is specific to TRegs, and suppress the proliferation of CD4+CD25- T cells.
