Isolation Of Human Regulatory T Cells

The purification of CD4⁺CD25^+bright regulatory T cells (T_Reg) has historically been a laborious, multi-step process, and is often complicated by contamination with activated (CD4⁺CD25^+dim) T cells. Now a rapid, simple method for TReg isolation is available. RosetteSep CD4⁺ T cell enrichment directly from whole blood is followed by manual (EasySep) or fully automated (RoboSep) positive magnetic selection of only high-expressing CD25^+bright cells. T_Reg purities of up to 90% can be achieved in 2-3 hours. The isolated cells express the FOXP3 transcription factor, which is specific to T_Regs, and suppress the proliferation of CD4⁺CD25^- T cells.
