Micro-HPLC For Proteomic Analysis

New 2D micro-HPLC system performs sensitive separation of complex peptide mixtures. The capillary-to-microflow system, co-developed by Shimadzu Scientific Instruments and the National Institutes of Health, separates complex peptide mixtures with resolution that complements present 2D gel electrophoresis. Highly ionic proteins and hydrophobic proteins can be analyzed on-line. Protein digests can be analyzed automatically by simply placing them into an autosampler. A sample from the cation exchange column is trapped on 6 mini trap columns (polymer packed reversed-phase columns with a rentention power equivalent to C4) for desalination, followed by reversed-phase analysis. System accessories include the AccuSpot MALDI plate spotter, which automates LC micro-fractionation, spotting and preparation for MS analysis.
